Introduction to Single Cell Omics
In the past decade, some of the important advances in molecular biology have been achieved through genome-wide and whole-transcriptome sequencing studies in individual cells. With the advancement of cell separation and next-generation sequencing, researchers no longer need to analyze the average signal from multiple cells in a population but can study DNA, RNA, protein, and chromatin at single-cell level. Single-cell genomics, epigenomics, transcriptomics, and proteomics studies have revealed differences in gene and protein expression even among cells that are genetically identical in the same tissue. At this stage, scientists have begun to combine multiple layers of information at single-cell resolution. These "multi-omics" techniques allow for a closer look at the variability between cells, more clearly identifying specific cells and their functions.

Applications of Single Cell Omics
We systematically characterize heterogeneity within tumors and the immune system using single cell analysis. We began with fluorescence-activated cell sorting analysis with surface markers and constructed the single cell library to detect genomic transcriptional profiles at single-cell level.
Single-cell RNA sequencing is of great significance for cancer immunotherapy research. The heterogeneity of expression in single cancer cells may be a factor limiting the therapeutic efficacy. Single-cell gene expression analysis can provide insights for prognostic markers and theoretical basis for combination immunotherapy.
Single cell sequencing has altered the ability to characterize circulating tumor cells (CTCs) and has been used to identify the metastatic potential of CTCs in cancer metastasis models to monitor abnormal signaling pathways for drug resistance prediction.
